Previously, we demonstrated that a daily consumption of milk containing Lactobacillus paracasei CNCM I-2494 (5 × 10(9) CFU) significantly reduced insulin resistance and serum cholesterol in overweight, insulin-resistant adults with type 2 diabetes mellitus (T2DM) and that the consumption of this strain increased the growth of the probiotic strain and the metabolic activity of the host (Maghrad et al., Diabetol. Med. 35: 576-584, 2018). Thus, the aim of this study was to confirm the effects of the consumption of milk containing L. paracasei CNCM I-2494 on insulin resistance in these subjects. In a randomized, double-blind, placebo-controlled trial, 80 overweight and obese insulin-resistant adults with T2DM (aged 60.8 ± 7.7 years; body mass index 29.5 ± 4.2 kg/m(2)) were assigned to consume their habitual milk product (placebo or skimmed milk, both containing 0.4 g/day of isocaloric lipids) daily for 12 weeks. Oral glucose tolerance test (OGTT) and a hyperinsulinemic-euglycemic clamp with stable isotope dilution were performed at baseline and after 12 weeks to assess insulin sensitivity. We also measured body composition by dual-energy X-ray absorptiometry.
